A headmistress is the woman who runs a whole school. Like headmaster, this is a British word, and it names the person in charge of the whole place, not just one class. It sounds even older than headmaster now. Many British schools prefer head teacher or the head, so they do not have to say if the person is a man or a woman. In American English people say principal for a man or a woman. You will still see headmistress in older books and in some private-school talk. Mistress here is the old partner of master: a woman in charge. It does not mean a secret girlfriend, even though that is how many first meet mistress.
