A question you ask to find out what films, shows, or events you can see. 'What's on at the cinema?' means 'What films are they showing?' 'What's on TV tonight?' means 'What programs are there?' You can also ask about a whole town: 'What's on this weekend?' means 'What events are happening?' It is a set question, so there is no object to move around. Though actually, be careful: 'What's on?' said to a person's face can also just mean 'What is happening with you?' — the situation tells you which one it is.
