Two everyday uses, and which one you meet depends on where you are. The main one: a person who is only somewhere at weekends. Think of a small seaside town where a few hundred people live all year and a few thousand arrive on Friday night — the locals call those arrivals weekenders, sometimes , often not, because weekenders push up house prices and vanish on Sunday. The word carries that faint edge of "not really one of us". The other use, especially in Australia and Britain, is for the things belonging to a weekend away: a weekender bag is a small bag holding just enough for two nights, and in Australia a weekender can be the little holiday house itself. In some places people also call a short trip a weekender — "we're doing a weekender in Tainan". So: a person, a bag, a cottage, or a trip, all hanging off the same idea of two days away.
組成weekend (Saturday and Sunday together) + -er (the ending that makes a doer or a thing associated with — like teacher, or like two-seater). weekend itself is week + end, the end of the week.
