To stop playing around and start working hard, usually because you have to — exams are coming, the deadline is close. You knuckle down, or knuckle down TO the work. Informal but perfectly polite; teachers and parents love it. This probably comes from the old game of marbles, where you put your knuckles down on the ground to shoot.
