Agitation usually describes a nervous, upset feeling that often shows on the outside - shaking hands, a shaky voice, pacing around. 'He spoke with agitation' means he seemed anxious and upset while speaking. The word is also used for organized public pressure for change, like political agitation, when people campaign strongly - sometimes noisily - for something to change.
